"Artificial Hematopoiesis" Research Makes New Progress

Japanese researchers have successfully used stem cells to grow red blood cells that can carry oxygen. On this basis, it is expected that a large number of red blood cells will be cultivated for blood transfusion to help the medical system relieve blood pressure.

The red blood cells of mammals have no nucleus and cannot reproduce themselves, which makes severe anemia patients currently only treated by blood transfusion. Large-scale laboratory cultivation of red blood cells has always been an important topic in Japan.

Although mature red blood cells cannot replicate themselves, the "semi-finished product" in the development process-red lineage progenitor cells have the ability to replicate. Researchers at Kyoto University in Japan reported in the online version of the new American journal "Stem Cell Report" that they found that two genes play an important role in the replication and maturation of erythroid progenitor cells.

The researchers introduced these two genes into induced pluripotent stem cells and embryonic stem cells, and successfully cultivated erythroid progenitor cells that could replicate and proliferate almost infinitely in the laboratory, and successfully differentiated them into mature red blood cells.

Most of the differentiated red blood cells are embryonic hemoglobin, which is different from adult hemoglobin, but the researchers confirmed that these hemoglobins have oxygen carrying capacity and can circulate in experimental mice after blood transfusion.

The researchers said they will further study how to differentiate erythroid progenitor cells into red blood cells containing adult hemoglobin, with a view to ultimately improving the blood supply tension in Japan.

Stem cells are "primary cells" that can develop and differentiate into different cells. Embryonic stem cells come from embryos and have the strongest differentiation ability. Induced pluripotent stem cells (iPS cells) are cultivated by "reprogramming" mature cells, and also have strong differentiation potential.
